Ipriflavone May Prevent Postmenopausal Bone Loss.

CHICAGO -- Ipriflavone, a synthetic isoflavone, can prevent postmenopausal bone loss and may be particularly beneficial in combination with hormone replacement therapy, Dr. Eric J. Bieber said at a conference on alternative and complementary medicine sponsored by the University of Chicago.

In one study, 56 women who had low vertebral bone density and were within 5 years of menopause were randomized to receive ipriflavone, 600 mg/day, or placebo. All participants also took 1,000 mg of elemental calcium daily.

At the conclusion of the 2-year study, a significant loss in vertebral bone density of 4.9% was seen in the women taking calcium alone. There were no significant changes in bone density in the patients treated with ipriflavone, a dietary supplement that's widely available in health food stores in capsule form (Menopause 5[1]:915, 1998).

Biochemical markers of bone metabolism and vertebral bone density also were evaluated in another longitudinal, 2-year study that compared calcium supplementation alone (500 mg/day) with three combination regimens: standard-dose ipriflavone (600 mg/day) plus calcium; low-dose conjugated estrogens (0.3 mg/day) plus calcium; or low-dose ipriflavone (400 mg/day) plus low-dose conjugated estrogens and calcium.

In the women given calcium alone or in combination with estrogen, there were no changes in urinary excretion of hydroxyproline or plasma osteocalcin levels, markers suggestive of excess bone turnover. Significant decreases in vertebral bone density also were observed in these patients (Maturitas 28[1]:75-81, 1997).

In the women given ipriflavone with calcium and those also given estrogen, urinary hydroxyproline excretion decreased significantly as did vertebral bone density. There were no changes in plasma osteocalcin ...
