2003 APR 16 - (NewsRx.com & NewsRx.net) -- A DNA vaccine encoding the ORFF gene conferred protective immunity in mice infected with Leishmania donovani, according to a report from India.
"The gene ORFF is part of the multigenic LD1 locus on chromosome 35 that is frequently amplified in Leishmania. The function of ORFF is unknown," said B. Sukumaran and colleagues, Jawaharlal Nehru University.
The researchers cloned "the gene encoding ORFF ... into a eukaryotic expression vector downstream to the cytomegalovirus (CMV) promoter. BALB/c mice were injected intramuscularly with ORFF DNA and challenged with L. donovani promastigotes."
The mice mounted both humoral and cellular immune responses against ORFF after vaccination with ORFF gene. This "provided significant level of protection against challenge with L. donovani," reported Sukumaran and colleagues.
