2002 MAY 29 - (NewsRx.com & NewsRx.net) -- by Michael Greer, senior medical writer - Researchers in Germany say that they have found a way to improve the benefits of idiotype vaccination for multiple myeloma patients.
"Idiotype (Id) vaccination provides an innovative treatment modality against B-cell malignancies," explained Dr. Matthias Zeis and colleagues at University of Kiel and the University of Leipzig. "In multiple myeloma patients, however, the antitumoral potential of this immunotherapeutic concept is limited."
The addition of interleukin (IL)-2 to the postvaccination regimen enhanced tumor regression and survival in mice, Zeis and coauthors found.
The researchers injected Balb/c mice with 1-5x10[superscript]5 cells expressing an immunoglobulin (Ig) idiotype protein specific to HOPC myeloma cells. Animals who were subsequently given either Flt3 ligand (Flt3-L), IL-2, or Ig[superscript]HOPC alone did not realize significant benefits from treatment, they said.
Myeloma-specific Ig protein with IL-2 produced complete responses in 27% of the study animals, while Ig[superscript]HOPC with Flt3-L induced tumor rejection in more than 40% of treated mice. However, the combination of all three agents enabled more than 80% of study animals to survive disease-free for at least 6 ...
