1902 MAR 27 - (NewsRx.com & NewsRx.net) -- by Michael Greer, senior medical writer - Genetic therapy for HIV targeting multiple viral functions may be an effective treatment modality, researchers in the United States argue.

"Compared with single agents, combination antilentiviral pharmacotherapy targets multiple HIV-1 functions simultaneously, maximizing efficacy and decreasing chances of escape mutations," explained Dr. David S. Strayer and colleagues at Thomas Jefferson University in Philadelphia. "Combination genetic therapy could theoretically enhance efficacy similarly, but delivery of even single genes to high percentages of hematopoietic cells or their derivatives has proven problematic."

Gene delivery vectors based on the SV40 virus may overcome these limitations, Strayer and coauthors said.

The researchers tested the efficacy of six recombinant SV40 vectors in delivering therapeutic transgenes and controlling HIV infection in vitro. Vectors based on SV40 have demonstrated a high degree of efficiency in gene delivery, they noted.

Cells from a human lymphoma line were protected against HIV infection after treatment with any of the six transgene vectors, alone or in combination, study data showed. However, only cells that expressed multiple transgenes after ...
