2001 DEC 26 - (NewsRx.com & NewsRx.net) -- by Michael Greer, senior medical writer - Researchers in the United States have found a way to enhance the performance of DNA vaccines based on the HIV pol gene.

"A synthetic gene consisting of the reverse transcriptase (RT) and integrase (IN) domains of human immunodeficiency virus type 1 (HIV-1) pol was constructed using codons most frequently used in humans," explained Dr. Danilo R. Casimiro and colleagues at Merck Research Laboratories in West Point, Pennsylvania, and the Veterans Affairs San Diego Healthcare System in La Jolla, California.

Proteins translated from this humanized form of pol induced potent immune reactions in animals, the researchers said.

Mice treated with humanized pol-based vaccines had high levels of T cells targeting the Pol protein, according to their report. Activated Pol-specific T-cell populations included both CD4[superscript]+ and CD8[superscript]+ cells.

A vaccine expressing synthetic pol coupled to a human tissue plasminogen activator leader sequence induced similar responses in rhesus macaques, study data showed. Circulating Pol-specific T cells in vaccinated animals rose to levels close to those seen in human HIV patients.
