Tamoxifen Reduces Incidence In Women With Inherited BRCA2, Not BRCA1.

2001 DEC 6 - (NewsRx.com & NewsRx.net) -- The drug tamoxifen reduces the incidence of breast cancer in healthy women who carry BRCA2 gene mutations that predispose them to the disease but does not reduce breast cancer incidence in healthy women with predisposing mutations in the BRCA1 gene, according to an article in the November 14, 2001 issue, of The Journal of the American Medical Association, a theme issue on human genetics.

Mary-Claire King, PhD, of the University of Washington, Seattle, and coauthor Bernard Fisher, MD, Scientific Director, National Surgical Adjuvant Breast and Bowel Project, Pittsburgh, presented the results of the study at a JAMA media briefing on genetics.

King and colleagues analyzed 288 women who were taking the drug tamoxifen or taking placebo and developed breast cancer during the Breast Cancer Prevention Trial (BCPT) between April 1, 1992, and September 30, 1999. The BCPT is part of the National Surgical Adjuvant Breast and Bowel Project. The BCPT recorded the incidence of breast cancer among those receiving tamoxifen versus the incidence among those receiving placebo. The researchers tested the study group for BRCA1 or BRCA2 mutations, finding that 19 women (6.6%) had one of the two mutations.

The researchers found that tamoxifen reduced the incidence of breast cancer by 62% in healthy BRCA2 mutation carriers, but did not reduce breast cancer incidence in healthy women with BRCA1 mutations. (Of 8 women with BRCA1 mutations who developed breast cancer, 5 were receiving tamoxifen and three were receiving placebo. Of 11 women with BRCA2 mutations who developed breast cancer, 3 were receiving tamoxifen and 8 were receiving placebo.)

According to background information in the article, the BCPT (which involved 13,338 women) demonstrated a significant reduction (49%) in breast cancer incidence among women who took tamoxifen. The benefit was evident in both women with no family history of breast cancer and those with mothers, ...
