Saltmarsh, staff medical writer - Mixing the diphtheria-tetanus-pertussis-inactivated polio vaccine (DTaP-IPV) with the Haemophilus influenzae type b (Hib) tetanus toxoid conjugate just prior to administration makes little overall difference compared with administering the two vaccines at separate sites at the same time, according to a new report in Vaccine.
N. Knutsson and associates working in Sweden studied 400 infants randomized to receive either DTaP-IPV mixed with Hib (DTaP-IPV/Hib (mix)) or DTaP-IPV given with Hib at separate injection sites (DTaP-IPV+Hib (sep)) at three, five, and 12 months of age.
Infants who received DTaP-IPV/Hib (mix) experienced fewer local reactions, but no serious vaccine-related adverse events occurred in either group, reported Knutsson and coworkers. Serum samples taken 28-45 days after the second and third vaccinations showed that Hib antibody titers were similar in the two groups after the second injection but lower in the DTaP-IPV/Hib (mix) group than in the DTaP-IPV+Hib (sep) group after the third injection (geometric mean 6.1 vs. 10.4 (micro)g/ml).
The mixed-vaccine regimen also caused somewhat lower diphtheria toxin antibodies (5.9 vs. 7.7 IU/ml after the third injection) but higher tetanus antibodies (3.9 vs. 2.5 IU/ml after the third injection) than the separate-vaccine regimen, noted Knutsson and associates.
