Saltmarsh, staff medical writer - Human polymorphic epithelial mucin (MUG 1) shows efficacy against several MUC1-expressing tumor cell lines and may be ready for further refinement in clinical trials in breast cancer patients, according to a new report in the International Journal of Cancer.

MUC1 is a glycoprotein expressed on glandular epithelia and overexpressed and hypoglycosylated in adenocarcinomas, explained F.G.M. Snijdewint and colleagues in the Netherlands.

"The objective of our study was to determine whether humanized MUC1 monoclonal antibodies (MAbs) and antibodies (Abs) induced by vaccination of breast cancer patients with MUC1 peptides can effect an antibody-dependent cell-mediated cytotoxicity (ADCC)," sald Snijdewint and coworkers.

The researches set up an in vitro assay using the breast tumor cell line ZR-75-I as the target and peripheral blood mononuclear cells (PBMC) of health donors as effectors, along with various MUC1 MAbs, to test efficacy.

They found that the humanized MAb HuHMIFG-1 induced cell-mediated cytotoxicity in nine MUC1-expressing tumor cell lines - including three bone marrow-derived cell lines - as well as two MUC1-transfected cell lines, to different degrees.

The PBMC from healthy donors exhibited large variations in killing capacity, and NK cells were the essential effectors for the MUC1 Ab-dependent killing, reported Snijdewint and team. Furthermore, plasma samples obtained from breast cancer ...
