by N.R. Saltmarsh, staff medical writer - The latest generation of adenovirus vectors are effective even in animals previously immunized with the same adenovirus serotype, suggesting their efficacy for humans with preexisting immunity, researchers in Italy have found.
Although helper-dependent adenoviral vectors have shown excellent gene expression in animal models, as well as limited toxicity after systemic delivery, they have a potential drawback, preexisting immunity to adenovirus, which affects a large proportion of humans.
"In this study we performed intramuscular delivery of helper-dependent vectors carrying mouse erythropoietin as a marker transgene," reported D. Maione and colleagues. "We found that low doses of helper-dependent adenoviral vectors can direct long-lasting gene expression in the muscles of fully immunocompetent mice."
When the researchers used the latest-generation helper-dependent backbones, which replicate and package at high efficiency during vector propagation, 100% of mice showed sustained gene expression four months later.
