by N.R. Saltmarsh, staff medical writer - Adding keyhole limpet hemocyanin (KLH) to dendritic cell vaccines can make them work harder. That translates to potent immune effects and rejection of established tumors in vivo, according to a new study published in Cancer Research.
K. Shimizu and colleagues in the United States evaluated the effect of KLH, a foreign helper protein, on tumor lysate-pulsed dendritic cells and peptide-pulsed DC immunizations in vivo.
"Besides being used as a 'surrogate antigen' in approaches to measure immunological response in cancer patients, KLH is also an immunogenic carrier protein to elicit T-cell help," explained Shimizu and associates.
They found that DCs pulsed with both KLH and tumor lysate enhanced immunity and rejection of established melanoma metastases in vivo. Cure rates and overall survival indicated that interleukin 2 further augmented this effect.
Adding KLH to DC immunizations enhanced tumor-specific T-cell production of interferon-gamma (IFN-gamma), reported Shimizu and team.
