by N.R. Saltmarsh, staff medical writer - A vaccine that includes the carbohydrate antigen globo H stimulates immune reactivity in patients with metastatic breast cancer.
Globo H was a likely vaccine candidate because it is commonly found on breast cancer cells, noted T. Gilewski and associates at Memorial Sloan Kettering Cancer Center in the United States.
"The objectives of this trial were to determine the toxicity and immunogenicity of three synthetic globo H-keyhole limpet hemocyanin conjugates plus the immunologic adjuvant QS-21," reported Gilewski and coworkers.
They vaccinated 27 metastatic breast cancer patients five times each with globo H plus QS-21. The vaccine was well-tolerated and all three formulations stimulated a similar immune response, namely immunoglobulin M (IgM) and, to a much lesser extent, IgG antibodies.
In 16 patients, IgM antibodies bound to MCF-7 tumor cells, but only a few patients showed IgG response, the researchers said ("Immunization of metastatic breast cancer patients with a fully synthetic globo H conjugate: A Phase I trial," Proc Natl Acad Sci USA, 2001;98(6):3270-3275).
