N. Kanesa-Thasan and colleagues at the Walter Reed Army Institute of Research tested the vaccine formulations in 40 volunteers with no immunity to flavivirus, in a randomized, controlled, double-blind study.
Five volunteers each were vaccinated with a single subcutaneous dose against dengue-1, dengue-2, dengue-3, or dengue-4; 10 were given a tetravalent vaccine with all four viruses, and 10 were given the vaccine vehicle alone.

All dengue-2, dengue-3, and dengue-4, and 60% of dengue-1 vaccine recipients developed neutralizing and/or immunoglobulin M antibodies. Dengue-3 and dengue-4 subjects were viremic between days 7-12, while dengue-1 and dengue-2 subjects were rarely viremic. All tetravalent vaccine recipients were viremic with dengue-3 virus and developed neutralizing antibodies to dengue-3.
