by N.R. Saltmarsh, staff medical writer - A single injection of diphtheria-tetanus-acellular pertussis (DTPa) and Haemophilus influenzae type b conjugate (Hib) vaccine causes fewer local and systemic reactions than diphtheria-tetanus-whole cell pertussis (DTPw) and Hib vaccines given separately, say researchers working in Spain.
J. Aristegui and colleagues evaluated the safety profiles of the two vaccine delivery approaches in 18-month-old children previously given three doses of DTPw and Hib vaccines.
"Children were randomized to receive DTPa/Hib (group 1; n=111) or DTPw + Hib (group 2; n=105) and were followed for up to 30 days post-vaccination," reported Aristegui et al. "All children received oral polio vaccine concomitantly. Local and general symptoms were recorded by parents on diary cards."
Parents of group 2 children reported significantly more local and systemic reactions than those in group 1. These included pain at the injection site (40% in group 1 versus 89% at the DTPw site in group 2), fever (10% for group 1 versus 56% for group 2), and fussiness (28% for group 1 and 71% for group 2).
In addition, parents reported lameness that resolved in one to five days for 21 group 2 children but only one group 1 child ("Reactogenicity and safety of DTPa vaccine and Haemophilus influenzae type b conjugate vaccine (Hib) ...
