by N.R. Saltmarsh, staff medical writer - Vaccination with the carbohydrate antigen globo H generates immunoglobulin M (IgM) antibody binding to tumor cells in patients with metastatic breast cancer, Memorial Sloan-Kettering Cancer Center researchers reported.
As a common constituent of breast cancer cells, globo H was considered an ideal vaccine candidate.
"The objectives of this trial were to determine the toxicity and immunogenicity of three synthetic globo H-keyhole limpet hemocyanin conjugates plus the immunologic adjuvant QS-21," reported T. Gilewski and colleagues.
They vaccinated 27 patients with metastatic breast cancer five times each ("Immunization of metastatic breast cancer patients with a fully synthetic globo H conjugate: A Phase I trial," Proc Natl Acad Sci USA, March 2001;98(6):3270-3275).
In most patients, the globo H vaccine stimulated production of IgM antibodies, and in 16 patients these antibodies were able to bind to MCF-7 tumor cells. Researchers observed no side effects and no differences among the three formulations. Only a few patients produced IgG antibodies.
